Synthetic D-retro-inverso peptide designed as a selective senolytic research tool targeting the FOXO4–p53 interaction.
FOXO4-DRI is a synthetic peptide engineered to disrupt the interaction between the transcription factor FOXO4 and the tumor suppressor p53 in senescent cells.
Constructed as a D-retro-inverso isomer (mirror-image D-amino acid residues in reversed sequence), it was published in the journal Cell in 2017 by de Keizer and colleagues as a research tool for senolytic biology.The preclinical research literature has characterized FOXO4-DRI's ability to selectively induce apoptosis in p16-positive senescent cells while sparing non-senescent cells in cell-culture and animal-model contexts.
Reported reductions in senescent-cell burden of roughly 25–30% within 10 days of dosing in animal models have made it one of the most cited senolytic peptide tools.Due to the synthesis complexity of all-D-amino-acid peptides, FOXO4-DRI is priced at a premium relative to conventional peptide synthesis products.
